Power Of Dream Ventures "PWRV" premier technology acquisition and development company operates through it wholly owned Hungarian subsidiary, Vidatech. Through Vidatech, we aim to provide pro-active support for ideas, research, start-up and expansion-stage technology companies having rights to technologies or intellectual properties which we believe to be potentially commercially viable, by offering a range of services designed to encourage and protect the continuing development and eventual commercialization of those technologies. PWRV focus is on technologies and technology companies based in the Republic of Hungary. PWRV strategy is to acquire majority  interests in technologies through, among other things, direct investment in start-up and expansion stage technologies and technology companies; cooperative research and development agreements with such companies; direct licensing agreements; joint venture arrangements; or, direct acquisition of technologies and intellectual properties.

PWRV recently announce the signing of an Investment Agreement with Genetic Immunity (www.geneticimmunity.com), one of the leaders of therapeutic vaccine product development. Cooperation between Genetic Immunity and PWRV dates back to 2009, when PWRV received rights to invest in GI and to issue press releases for the Company. With today's signing of the Investment Agreement over two years of close collaboration has finally come full circle. Founded in 1998, Genetic Immunity is a leading biopharmaceutical company leveraging its proprietary nanomedicine technology platform to develop Therapeutic Vaccines. DermaVir, the Company's Lead Therapeutic Vaccine Candidate, has been developed for the treatment of HIV/AIDS, with recently completed Phase II clinical trial showing clinically significant median 70% viral load reduction in vaccinated patients as compared to placebo without systemic side effects. Such excellent safety and immunogenicity were seen in up to seventy HIV-infected people treated with DermaVir demonstrating the human Proof of Concept for the technology platform.

Genetic Immunity Technology 

Until today plasmid DNA based vaccines have been proven safe but poorly immunogenic in human subjects. Genetic Immunity developed several technologies to improve the immunogenicity of DNA-encoded antigens by delivering them to Dendritic Cells.

Genetic Immunity's Therapeutic Vaccine Platform: three core proprietary technologies designed to address the challenges of 1) appropriate antigen composition, 2) delivery, targeting, expression and presentation in dendritic cells and 3) clinical development and commercialization:

  • ANTIGENeering: Rational design of a single plasmid DNA molecule (pDNA) expressing multiple safe antigens to induce a multi-faceted immune response.

  • Nanomedicine Formulation: Proprietary pathogen-like nanoparticle formulation for use with pDNA antigen for effective delivery and antigen presentation by dendritic cells. Dendritic cell targeting by the nanomedicine formulation is antigen independent. Excellent safety and stability data from preclinical and clinical studies. Technology appropriate for commercial scale production.

  • DermaPrep: CE Marked Transdermal Delivery Device for Vaccines and Liquid Formulations:
    • Initially designed for topical delivery of antigens to dendritic cells and to replace ex vivo dendritic cell-based immunizations. Delivers the nanomedicine to the dendritic cells of the lymph nodes via epithelial Langerhans cells.
    • DermaPrep also has potential for use outside of vaccines (e.g. topical delivery of liquid /gel forms of proprietary or generic therapeutics; esthetics/cosmetics)

 

Genetic Immunity Clinical Trials

Genetic Immunity's DermaVir is our candidate Therapeutic Vaccine designed for HIV-specific immune reconstitution. The Antiviral Mechanism of Action is based on inhibition of viral replication by cytotoxic killing of HIV-infected cells. Currently, there are no approved immune therapy or vaccine products on the market for HIV/AIDS.

DermaVir Phase II. results

It is a concern that vaccinations paradoxically increase viral load. Therefore, GIEU006 was designed to evaluate safety of repeated DermaVir immunizations in antiretroviral-naïve subjects and also test the immunogenicity and antiviral efficacy to guide later development. DermaVir includes a pDNA expressing an HIV (Clade B) virus-like particle formulated into a topically administered synthetic nanomedicine.

36 HIV-infected adults (HIV-RNA: 5,000 to 150,000 copies/mL and CD4: =400 cells/mm3) were randomized to receive one of three DermaVir doses (0.2, 0.4 or 0.8 mg of pDNA) or placebo at weeks 0, 6, 12 and 18. Standard parameters and HIV(gag)-specific precursor T cells were quantified through week 24. ITT was analyzed by non-parametric statistical tests, the treatment effect with Hodges-Lehmann estimator.

Baseline characteristics were comparable across groups (median age: 38 years; CD4: 506 cells/mm3; HIV-RNA: 20,250 copies/mL; 83% Caucasian; 97% male). No subject stopped vaccinations due to an AE and only one subject initiated ARV. No AE =Grade3 occurred, grade 1 and 2 incidences were similar across groups, and only one Grade2 AE was possibly-related to the vaccine. Anti-dsDNA decreased, ANA and CD4 counts did not change and HIV-RNA did not increase. Based on secondary analyses, the 0.4 mg group was superior to others. In this group the HIVgag-specific precursor T cells increased from 5,055 to 9,978 cells/millionPBMC (P=0.07). The median log10 HIV-RNA decreased from 4.5 to 4.0, significantly different from the placebo (P=0.045). The treatment effect compared to placebo at 95% confidence interval was -0.23 (-0.70; 0.09) for log10 HIV-RNA.

DermaVir did not increase viral load nor reduced CD4 counts and was as safe as placebo. Its antiviral and immune-boosting effects in the challenging antiretroviral-naďve setting support further development. The 0.4 mg dose emerged as the vaccine candidate for the early treatment of HIV infection.

For example, an article out today titled 'Ozone layer faces record loss over Arctic' contains many reference points such as names of geographic locations, organizations, cities and people. With iGlue any one of these entities can be annotated. Users can add images, videos, maps, comments and more to 'Geneva, or 'World Meteorological Organization' or 'ozone layer' to provide value added relevant information on them without leaving the page to search for more insight.
